DKW Medium โ€“ Driver and Kuniyuki Walnut Medium

 

DKW Medium, short for Driver and Kuniyuki Walnut Medium, was initially formulated for the micropropagation of walnut using nodal explants. Today, it is widely used across various plant species due to its unique nutrient profileโ€”characterized by lower nitrogen and higher sulfate concentrationsโ€”which may positively influence shoot regeneration and overall plant development.

How to Prepare DKW Medium (1 Litre)

 

Follow these steps for reliable media preparation:

 

In a beaker, pour 800 mL of distilled water.

 

Add the following:

 

5.36 grams of PCTโ€™s DKW Medium powder

 

30 grams of sugar

 

1โ€“2 mL of PPMโ„ข (Plant Preservative Mixture)

 

Top up the solution to a final volume of 1000 mL.

 

Adjust the pH to 5.4โ€“5.8 using a pH meter and appropriate reagents.

 

Incorporate a gelling agent:

 

6โ€“8 g/L agar or 3โ€“4 g/L gellan gum, and continue stirring.

 

Autoclave the medium at 121ยฐC and 15 psi for 20 minutes.

 

Your DKW medium is now ready for use!
